Pharmaceutical company Janssen has declared the ineffectiveness of its vaccine against HIV named Mosaico.
The Johnson & Johnson company came to this conclusion on the basis of the third phase of clinical trials.
In 2007, scientists developed an HIV vaccine that consists of sets of mosaic antigen proteins.
They included fragments of the Env, Gag, and Pol viral proteins, which contained amino acid substitutions characteristic of the main strains of the virus.
This approach provides greater antigenic diversity of the vaccine.
In 2018, the Mosaico HIV vaccine successfully passed phases I and IIA of clinical trials, showing its safety and immunogenicity.
For that reason, scientists began to plan the next phases of research.
The third phase of clinical trials began in 2019 and ended in October 2022.
It was attended by 3,900 people at risk from Argentina, Brazil, Italy, Mexico, Peru, Poland, Puerto Rico, Spain and the United States.
Participants were given the vaccine 4 times over the course of a year.
UNSUCCESSFUL TRIALS
In January 2023, the pharmaceutical company Janssen, stated that the vaccine did not show its effectiveness in protecting against HIV infection in at-risk groups.
According to the results of the third phase of trials, the injections did not result in a significant increase in neutralizing antibodies compared with placebo.
At the same time, scientists did not find any problems with the safety of the vaccine.
The company is halting further research and is currently continuing to analyze the available data.
Officially, the results of the third phase have not been published.
The termination of the clinical trial of this vaccine comes amid the halt in August 2021 of the phase IIB trial of Imbokodo, another HIV vaccine from Janssen.
It also did not show significant results in protecting against HIV.
Mitchell Warren, executive director of the HIV Vaccine Development Coalition, said at least 5 experimental HIV vaccines failed to demonstrate efficacy in phase III trials.
"The current failure puts a big question mark over this area of research," Warren suggested.
The main problem is that, it is not known who else would agree to invest in this area of research after such results.
