THE DIARY GAME || 8th February 2023 || Morning Routine, Laboratory work ( DNA extraction and Quantification.)

I am very glad to share my diary game with you. I strongly believe you will enjoy the day with me. Hen come let's visualize and take you through my activities for the day.

Today happens to be Wednesday, the 8 days of January in the year 2023. This particular day which is midweek has a lot to say about workers. Most of the time, workers will be very weak because they have used most of their energy to work in the previous days. However, we still have to put in our maximum best in other to be able to achieve our ultimate goal. Fortunately for me. I was having the required energy for the work ahead today.

I was awake around 6:02 AM. And because I cook in the evening yesterday I was having cooking utensils to wash. I quickly brought them out to wash first. When I was done doing that, I then went to pick up the broom to go and do the normal sweeping of my room. I did that also in a hurry and then quickly pack my things to be able to arrange my place neatly. I did that in no time making my room look neat.

I then went to the bathroom room to take my bath. But before that, I picked my things including spoke and peel before going to fetch the water. After I was done, I sent it to the bathroom and showered. I returned quickly to finish up. What I did was to get my comb, get my pomade, and the mirror after using all these I kept them down, and wore my dress, and left. It was getting late hence I left to work.

In about fifteen minutes, I was at the workplace. Fortunately for me, I log in with my details and went to the lab to do bench cleaning. I took my time to be able to do all these. Right after that, there was work ongoing hence we have to start as quickly as possible since we were having DNA extraction to do. After doing all that I went to the store to bring out the extraction kit.

After bringing it up, I realize that is not the one I was supposed to bring because it has expired. I sent it to the disposable goods room and picked a new one for extraction. It took us a few minutes to be able to get all of them ready to be used. When they were ready, I set up the hood and started working on them. Unfortunately, they were more of it, we were having fifteen Samples to extract the DNA for.

We quickly brought out those plates and harvest the cells on the lysate buffer. We continue with the process and in about 4 hours, we were done with the extraction. I then have to pick my things so that I can do the normal was of doing them. It was left for us to do the quantification of the DNA samples. It took us a few minutes to prepare the working solution. When I was done, I picked it up and did the quibitting and fortunately out of the 15 samples, only two couldn't pass the quantity we would have loved to have.

In any case, we were using a broad range which means they are still high quantities in the sample. If we change to use the high sensitivity we might get a high yield per the extinction we did. We went ahead to go and rest. Because it was time for closing, we left and went to our various homes. When I go home I rested and took food. I then took my bath and went to bed.
