Nick Fury has been around since World War I , which he worked along side with Captain America and Bucky on several missions. On a one mission his left eye was several damage and due to lack of medical attention , it totally damage his eye.He did take a "Inifinity Formula" , which makes Nick Fury halt his aging process. But taking them them regularly to make him alive.Source here
