Religion is the main difference.
Pakistan is 96 percent Muslim.
India is 80 percent Hindu and 13 percent Muslim.
Ethnically, the populations are different.
Pakistan is 45 percent Punjabi, 15 percent Pashtun, 14 percent Sindhi, and ~25 percent from several other South Asian ethnicities.
India is 72 percent Indo-Aryan, 25 percent Dravidian, and 3 percent other.
Their languages are different.
Pakistan's languages are Punjabi (48 percent) and many others, Official languages Urdu (8 percent) and English.
India's languages are Hindi (41 percent) and many others, Official languages Hindi and 14 others, plus English as a subsidiary official language.

Interesting, in college I worked at a grocery store with a Pakistani man, he said in the U.S. Indians and Pakistanis are friends, but of course that couldn't be said for national relations in their home countries (-:
