There are three stages involved in spawn production in the order below.
i)Raising of pure culture.
ii)Preparation of mother spawn.
iii) Multiplication of spawn.
But in this post am going to talk about step 1(raising of pure culture.)
Preparation of the growth media.
Preparation of media is key the process of raising pure culture.
Commonly used media
- Potato extract.
- malt extract.
- corn meal.
Most of the media are available on make but can be prepared according to instruction given by the manufacturing company.
Preparation of potato dextrose sugar.
1)Wash the potatoes and slice them into very small pieces.
2)Then place them in boiling water. Per 1 liter of water in the flask add 200g and boil for 15 minutes.
3)Use muslin cloth to filter the potato broth.
4)Add 25g of agar and 25g of. dextrose and add water to increase the volume.
5)Sterilize the agar mixture and petri dishes in the autoclave.

- Then pour the media carefully into petri dish (about 20ml per Petri dish).
Keep the petri dish or test tube in one position so that the agar solidified.
In one day the agar slants can be used for pure culture.
The are two methods of raising pure culture
1)Tissue culture technique.
2)Spawns technique.
Tissue culture technique.
Get a fresh mushroom and sterilization it in alcohol. Cut off all the external parts and the internal basidiocarp.
Cut it longitudinally into 2 halves and bits.

Transfer the bits into the sterilised PDA or culturing media.
The petarepetri dish are incubated between 240C and 260C for 1 week.
Mycelium from growing edges is carefully transferred an other PDA and incubated for 2 or 3 more weeks obtain pure cultures.

Mushroom spawn can be prepared on any kind of cereal grains like wheat, jowar, bajra or rye and agricultural waste like corn cobs, wooden sticks, rice straw, saw dust and used tea leaves, etc.See Spawn substrate i.e. cereal grains should be free from diseases and cereal grains should not be broken, old, and insect damaged. Washed grains are then soaked in sufficient water for 20-30 minutes and boiled in a container for 20-25 minutes. Normally for soaking and boiling 20 kg of wheat grain, 35 liters of water is required.
